This can be a long task. The first step is to recruit an assassin in the city. Then you must send them on missions from the bird coup. They gain experience from each mission. Once they become an assassin, they must be assigned the Master Assassin of a Den. At that point they will have a memory you must complete so they can grow past level ten. At this point they must be sent on more missions until they reach level fifteen. Then you must complete a second memory which shows your Protege has grown into a true master assassin. At this point they become a Master assassin rank 5 and their den can no longer fall under Templar attack.
